Associate Scientist, Gene Expression Analysis - Gene Therapy

 

Summary of Position

This position will be responsible for the quantitative determination of gene expression magnitude and copy number pertaining to Gene Therapy constructs in cell-based and in vivo models.

Roles and Responsibilities

  • Isolation of mRNA from cell and tissue models
  • Design and evaluation of qRT-PCR and ddPCR probes
  • Selection and validation of representative stable control genes for each sample type
  • qRT-PCR-based and/or ddPCR determination of mRNA expression and gDNA copy number
  • Propagation of cell lines transfected or transduced with gene therapy targets
  • Measurement of target mRNA expression levels in naïve cell lines and tissues
  • Ensuring documentation of all work in an electronic notebook
  • Analysis and presentation of results to larger scientific group

Qualifications and Background Requirements

  • BS or equivalent in a scientific discipline, and a minimum of 2 years lab experience with RNA isolation and analysis, or MS or equivalent with 0-2 years of experience with RNA isolation and analysis
  • Successful isolation of high-quality mRNA from a variety of sources for the purpose of quantifying mRNA levels of endogenous and transgene targets
  • Evaluation of isolated RNA quality and quantity
  • Familiarity with mRNA target quantification techniques (e.g., qRT-PCR, ddPCR)
  • Knowledge of the design and validation of primers for qRT-PCR and/or ddPCR
  • Experience with qBase and ddPCR preferred
  • Ability to support multiple projects simultaneously
  • Able to function both independently and as part of a team
  • Demonstration of detailed record keeping and data documentation

Travel

  • Occasional travel to scientific conferences to present work or for training courses
